Scaling Up of Sugar Separations Made Simple, with Knauer’s Refractive Index Detector

The scaling up of the separations of sugars on HPLC can sometimes be a problem due to the high sensitivity of refractive index detectors (RID) and their limited dynamic range. In this poster, two sugars are separated at different concentrations to demonstrate the effectiveness of the extended dynamic range (EDR) option for Knauer’s AZURA RID 2.1L refractive index detector.

Different volumes (10 μL, 20 μL, 30 μL, 40 μL, 50 μL, 100 μL, 200 μL) of 40 mg/ml solutions of glucose and saccharose were measured with and without the EDR function, with the graph here illustrating how the EDR function has extended the gain of the detector by about 65%.

It is shown that when using the extension, better values of linearity and correlation coefficient can be achieved over a wide range.
